Hilton San Francisco Union Square. Booked King room, upgraded to microsuite (a little bigger than an ordinary room, not big enough to be a minisuite) at check in.

The Diamond Desk specifically (or more generally the method, time and place of making a reservation) has no bearing on upgrades. Upgrades, if any, are given by the hotel at the time of check-in. The reservation may have been made an hour, a month or 6 months before - the moment and point of check-in (and rooms available at that time) is what is relevant.

An increasingly rare suite upgrade for me:
Doubletree West Palm Beach Airport
Booked AAA King Rate. When I checked in the agent asked if I wanted a high floor or low floor and I said "a suite if you got it" and was told I'd already been upgraded to a junior suite ^

Booked King Deluxe Room
Got a King Deluxe Room
Said they have no suites available - looks like they might have pre-upgraded others as there were multiple levels of suites for sale as of a couple nights before check-in. Now all suites look to be sold out for the first night of our stay. First time that I haven't been upgraded as a Hilton Diamond and I even booked through Virtuoso.
EDITED TO ADD: Management was able to find a suite upgrade for our last night, but we were comfortable enough in the Deluxe room that we decided to stay there. There is only one sink in the Deluxe (versus 2 in the suites), but there is also a bar sink that we made work for us when we were getting ready in the mornings.
