Additional Driver bypass desk
I am traveling with my wife, and want to add her as an additional driver. Normally I travel alone and go right to my car. Can I add her at the check out booth or do I have to go to the Preferred Desk?

My guess is that if you're not a CHM, you'll have to go to the Preferred counter, as she'll have to fill out an additional driver form (including cc number, DL, and sig). Don't know about extra driver charge though... check your rate.
(CHM are entitled to free additional drivers, with no forms needed for spouses and co-workers)

I know that there is no charge, but I was hoping to avoid the line. But at least I am going into MSY at 11:30 PM. Shouldn't be too much of a line.

In my experience, no need to register a spouse/coworker as an additional driver.
Go to avis.com->Customer Service (top right corner)->FAQs->USA->Reservations->Rental Policies, Terms & Conditions:
The following individuals, if 25 years of age or older and in possession of a valid driver's license, may drive the car with the renter's prior permission and are exempt from Additional Driver fees:
Renter's employer (while on company business);
Renter's fellow employee (while on company business
Renter's spouse or life partner.
Disabled renters who have completed the non-licensed renter form
Late last year, we hit something in the road with a rental in my name and she wasn't listed as an additional driver. My wife filled out the accident form/signed it (and so did I as the renter) and they didn't question her being the driver.
It's also worth noting that Avis considers "life partner" someone with the same address on the driver's license. I've questioned them a few times on this (to be sure the policy hadn't changed, before my now-wife and I were married) but always get the same answer!

Thanks for pointing that out. The operative part is really the next paragraph:
A maximum of 2 additional drivers can complete an additional driver form* for a rental. To save time, you can print out this form, complete and bring it with you to the counter. Additional drivers who can drive the vehicle with the renter's permission (see Exceptions above) are not included in this requirement.
That means no counter or form for me!
