Passport requirement went into effect at the beginning of 2010. The law passed not long after 9/11, but the feds kept postponing the enforcement date until last year. If your friend from YYZ is going to cross into the US, he will need a passport too.
Windsor train station is not downtown, but a mile or two east. Not sure what transportion options there are from VIA station to downtown Windsor. I assume taxi would be quick and easy. You might check if Casino Windsor runs a shuttle.

Windsor VIA rail to downtown Windsor/tunnel is roughly 2 miles. Taxis are available at station (station is manned most of the time). A little too far to walk. Tunnel Bus runs underneath and drops off at Mariners' Church/Renaissance Center.
Other than the to-ing and fro-ing, I think I'd just pick your friend up at the train station and either do Windsor or Detroit and be done with it (I think I'm reading that you'll have a car), or have him take a cab across (in the interests of time).
