Unable to Modify Dates for Reservation Booked with Points

I have a week-long reservation for Comfort Hotel Tokyo Kanda for the 25th, but I need to check in on the 26th. I booked with points through Choice Privileges, but it looks like I'm unable to modify the dates because the hotel is fully booked. Should I cancel the reservation and rebook? I'm worried that the room won't get put back into the inventory for points bookings.

You can try calling and seeing whether a human representative can help you knock off the first day of the reservation, but its possible you might just be stuck. Whether a cancelled reservation goes back into points inventory or not is pretty inconsistent from program to program. If youre booking something you suspect might be a competitive reservation and you think theres risk of changes, its generally advised to book individual days; as long as theres no bulk pricing for a longer stay, theres no downside beyond a little extra time booking.
