4 days before flight departure and precisely the exact time the flight is scheduled for.
Lets say your flight is on Oct 4th at 2pm, you should be upgrading today(4 days before) at 2pm.

If it clears before the flight, you will get an email to confirm.
If it doesn't not clear before day of, you will be transferred to the airport list - check in at 24 hours to help your priority and then check with the AC staff when you get at to the airport to make sure you are on the list.

You don't know how many people were on the waitlist already ahead of you at 96h.
These include: anyone with eCredits who purchased a Y or B fare at any time before 96h, Super Elites on any fare who waitlisted before you starting 7 days before the flight, and any Elites who squeezed in before you.
At 24 hours things get more nebulous. It is not FCFS. Status, fare, check-in time, and the phase of the moon also play roles.

I just got Mrs DavidYYC an upgrade (as my nominee with no status) at exactly the 96 hour mark on AC 850 YYC-LHR.
I think (could be wrong though) that if there is no-one on the wait list, it is first come first served for E and their nominees to fight it out at the 96 hour mark.
The R number on this flight changed from R3 to R0 after the 7 day mark the back up to R1 then R2, before I upgraded.
If it was R1 when I did the request and there was another E who wished to upgrade themselves at the 96 hour mark, they would have lost out as my non status nominee got the last R seat at exactly the 96 hour point, he/she would lose out if their request came after mine.
There is no way that the system would know who wants an upgrade along with my nominee, until it is requested.

We are both waitlisted for an upgrade for the YYZ-HAV flight tomorrow. I just looked at the availability and there is 1 R space open. Now does not AC's programming grab that one seat into R and upgrade one and waitlist the other?

My suggestion is to cancel the waitlist and split your travel into separate PNR's (assuming your travel companion can be added as a nominee). If your nominee has no status, upgrade them first as you'll be more likely to have your upgrade clear at the gate than a non-status companion (of course seating arrangements may not be desirable by that time).

Yep, got it, gee, grabbed that 1 R spot and break up the file. Now the other party is waitlisted. Don't think there is anyone else on the waitlist as the R spot was there for more than an hour. It refreshes every 15 mins. ^
