This was answered by UA Insider earlier: if your MileagePlus number is in the reservation when you made it, it'll automatically be queued up into the CPU process.
But CPUs are not clearing automatically for many people for flights this week, as per many posts on this thread. A call to UA is definitely in order if R availability is showing and you're within the upgrade window (e.g. 96 hours for 1K), but not yet cleared.
