Remove the bedspread and then wash my hands.
if I am staying at a hotel with duvets instead of skanky bedspreads, I can skip this step.

I would say clean the T.V. remote control. This has been proven to be the filthiest item in the room and rarely if ever cleaned by housekeeping.It's a real disease spreader.

Check the bathroom to see how nice/bad it looks.
Check the clock radio to make sure it's not set to some early morning time.
Check to see how good the water pressure is in the shower.
Sit on the bed to make sure the mattress isn't lumpy.

I tend to walk around and check out the room in a not so orderly way (could be compared to a dog circling before he lays down). I really wish that I could just be happy/not care about what room I am in...but I always do. I almost always check the TV style and then the drawers to see where I would put my clothes if I ever stayed somewhere for more than one night.
It is rare that I go into a room and not think that I smell smoke. Especially in Manhattan.
Then I call downstairs and ask for foam pillows to replace the feather ones.

Go right to the window to check the view, which is very important, then check the bathroom - hoping it is large enough and has a good size tub. And usually we check to see if we have down pillows and if not, put in a call to Housekeeping! And oh yes, if there is the slightest whif of smoke I ask to change rooms.

Adjust the AC (usually it's too warm) and then look at the air filter to see if it's relatively clean (with dust/smoke allergies it makes a big difference). In 99% of hotels in which I have stayed (without requesting a clean filter in my res.), the air filter looked as if it had not been changed in months, if not years. (There seems to be enough research out there to support the assertion that a dirty air filter requires more energy to heat or cool the room; I've never understood why the hotels don't have monthly or bi-monthly programs to change these 50 cent filters more regularly.)
