Can someone tell me what the following mean "R0 X0 N3"? Thanks.

In what you'd like to know:
R0--No first class award tickets (commonly XF)
X0--No first class upgrade awards from paid business (commonly NF)
N3--Three economy award tickets available (commonly XY)

why do they sometimes put R0 if there are no first class award tickets and sometimes not write anything at all? If there a difference?

R0 indicates that there is a F cabin on the plane. If that isn't there, there isn't an F cabin!
