There was a thread on FT recently where we were tying to guess airport codes that were the same except for a rearrangement of the three letters, but there were some additional conditions. I remember guessing ANC to CAN.

I started the thread intending it to be about non-stop segments. Of course, individual segments could be joined to make a longer itinerary.

I’ve actually been working up a series of itineraries based on license plate letters for the Old Timers’ Airline and Airliner Quiz and Discussion ... the inspiration was seeing cars with BGM, BHM, BHX, and BLR in quick succession

Three non-stops that I've done - quickly come to mind:
LIS-LYS
LAS-LAX (already mentioned)
IAH-IAD (already mentioned)
PMI-PMO - exists, but I haven't flown that.
MSP-MSY - exists, I'm surprised MSP Economist didn't mention this.
Many ITIN possibilities.
e.g. LIS-...-LOS (also flown that).
Same letters but transposed:
DUB-BUD (haven't flown that)
SOF-SFO (no nonstops on that, haven't flown it either)
IST-ISL was a theoretical possibility earlier this year

Do you mean San Luis Obispo, CA? The airport code is SBP. Was it ever SLO? SBA-SBP would be nice but not sure if there was ever commercial service on that 72-mile distance. In that region, SMX (Santa Maria, in between the two) to SMF might be possible.

Long ago, you probably could have flown OKC-MKC. Now you'd need a charter. When the Thunder play a preseason game in Kansas City, I assume they fly this.
I forget if TWA ever served the Virgin Islands, but if so there could have been an STL-STT or -STX.
I've flown some IND-BWI and IND-DCA but never an IND-IAD.

before the code changed to BWI (~1985), you could have done BAL-BDL (and very likely BAL-DAL in the 1960s)

Nobody cares about connecting flights. There become thousands of combinations then. The only interest is nonstops.

I’m sure you’re familiar with puzzles in which the object is to go from one word to another by changing one letter at a time, such as
CAT -> COT -> COG -> DOG
That got me wondering how many “change one letter” flights FTers have done, and whether anyone has done a multi-flight chain itinerary a la SAN-JAN-JAX.
