I hate FRA transfers and I hate flying away from my destination to get there, so I'd take the KUL transfer even if its means no A380. I like KLIA a lot.

The latter would be my initial thought.
I don't know about TG's A380 service, but my EK A380 flight was fantastic, much better than the A340 and 777 legs I flew, even though on the same airline.

I have flown on the Air France A380 before and do not see the big deal. In fact, the flight also happened to have some of the worst turbulence I had experienced in about 30 years. The food on Air France is sometimes quite nice, so that is a plus. Be aware though that Air France has a habit of cancelling or severely delaying flights. That's exactly what happened to us. There were passengers who told us this had been the third time they had had their flights cancelled due to technical problems. We all lost at least a day.
I have never flown Turkish Airlines so I cannot comment there.
My initial response, based on what I have experienced, is that you should rather go for option 1 as the travel time will be less. Seeing as you fly Business Class and have not experienced the A380, I would rather recommend you book yourself on any A380 if it means not having to first fly in the wrong direction.

I would take the second option (TG A380) for a few reasons:
1. After a long flight, I just want to go to my destination. I prefer a short flight before a long flight, not afterwards.
2. TG's 380 is amazing, especially in business. I flew it from BKK to FRA in December and it was one of the best business class flights ever.
3. As an FT'er, longer flights are better since they give me more miles
