Where get an info about route
HI
I plan to write an application, which would do some calculation about number of miles, that should be earned for particular segment. However to do it, I need info like:
• Is the flight domestic?
• Is the flight within the same continent?
• Is the flight within one area (for example European Union)?
• What is the miles distance, between departure and arrival city?
Do you know any source, that I could use here?

Have a look at openflight data: http://openflights.org/data.html. They have most of the routes, airlines and airports. With a bit of crunching you can get all the information you need.

Thank you. It is a good starting point.
Now the challenging part is to find is the flight: domestic, within the same continent or area...
Some miles programs have different calcualtions for different type of routes..

Where get an info about route
The great circle mapper (Google it since I'm too lazy to look it up on my phone) has been 99% accurate anytime I have checked against award miles. You may be trying to reinvent the wheel.
