Which airlines do you currently park your miles?
The best way and least miles are not necessarily the same thing. It can be a quality (re:service) vs, quantity (re: miles) thingy.
Are you mainly asking for fewest miles in C from SFO to SIN?
Please keep in mind, not every mile is created equal. Miles in certain FFPs sometimes hold higher redemption value than others. And the value is in the eye of the beholder and/or hoarder.

OP has specific Q - least miles in C to SIN.
AFAIK, the least would be on BA miles, using JL via NRT or CX via HKG. It would cost 50k miles (BA) each way.
If you do a transfer from Amex to BA before 7/31, you just need 67k MR for the R/T.

Miles not the only concern
Don't forget that each airline program or airline used to get you there also have different fees that you must pay out of pocket and the fees varies ALOT. Some fuel surcharges with some airlines are very high while others are not. So its not just the miles.

Do you have hundreds of thousands of miles in every program available? Or are you just curious? Outside of those two scenarios we're going to need a lot more info.

How is the SFO-SIN trip in business, not 100,000 in BA miles? from

@bbkenney -- It depends where you need to go. thepointsguy just posted on his blog about the 50% transfer bonus on Amex to BA or DL and gave several travel scenarios.
I'm actually thinking about SFO-SIN this November. The availability on CX is probably better than JL; there are two CX SFO-HKG flights per day and 8 HKG-SIN flights per day. A 100k BA miles RT in C is an excellent deal and the taxes are around $300 USD.

I really like Cathay, foods and services are great.
BUT Singapore airline is one of the best.
AA miles from SFO to HKG to SIN with Cathay (tax is about $60 per person - plus 70,000 miles (Coach) or 110,000 miles (Business))
