yes if you are paying when you check out, regardless of payment method.
no if it a "direct bill" arrangement e.g. you just sign a sheet when you come in but never see/pay a bill. example of this is airline flight crews. A hotel may still give you access in these circumstances, but they are not required to

Well it depends on the brand, first of all. Not all Marriott/SPG brands provide free club access to elites.
In addition to the direct billing exception, there have also been some reports of corporate rates that expressly do NOT include elite benefits.
