Problems With Using AmEx Poins For CO Upgrades?
I am considering taking advantage of AmEx's offer of 25% bonus points if you transfer CO miles to your AmEx account. My question: is it a procedural pain in the butt to use these AmEx points as opposed to CO miles to upgrade (say to first class from coach)? It is a seamless thing to do with CO miles, and would like to know if I am opening myself up to a major runaround. Thank you.

it's not a problem at all. as far as CO is concerned, they have no way of knowing where the points came from, unless they go looking at your statements to see the AMEX transfer.
in some ways it is actually easier to book awards. if you don't have the miles in your account yet, tell the agent that you are transferring miles from AMEX. they will allow you to hold the space and note in the record to not cancel the space "pending AMEX points transfer".

Pardon my stupidity, duxfan, but based on the implications in your post, I must transfer the once CO miles (now AmEx points) back into CO miles in my OnePass account to use them, no? In other words, the CO booking agent can not yank the AmEx points from my AmEx account, true? Thank you.

Correct. You transfer the MR points into OP miles. Beware, they can never go back!
As mentioned, the best thing to do is find the flights/fares, etc that you want to travel on, tell them to "hold" the reservation pending transfer.
Then initiate the transfer, about 3 - 5 biz days, and book the tix.

Remember that you can use 6000 MR points to get a $99 companion pass on CO and on other airlines as well. I thought about transferring some points to get that extra 25%, but I think I'll leave them there and use them for the companion passes instead.
Just a thought - - consider all your options!
