Yes, the seats will go out empty if no one purchases them or uses other upgrade instruments (such as miles or Global Upgrade Certificates) to upgrade to them. Only Comfort+ seats are complimentary upgrade eligible on long-haul international flights (with a Delta One cabin) while Premium Select and Delta One seats are not. There's no point in repeatedly checking flights for comp upgrades as it won't happen.
