Generally speaking, the name, the DOB and the Known Traveler Number in the PNR should match the passenger's trusted traveler's profile in order for the PreCheck designation to appear on the boarding pass. Since your wife's KTN is associated with her NEXUS membership - I assume that she does not have an stand alone PreCheck KTN - all changes must be made through NEXUS. Perhaps you can have her name updated at a Global Entry Enrollment Center (NEXUS and Global Entry are both administrated by the CBP, PreCheck by the TSA).

FWIW: not in my experience.
I have flown several times recently with a ticket under my now legal name, not remembering when I booked it that my Nexus card (hence TT number) had my old name. I entered my TT number as usual, got Pre-Check as usual. The first three times I flew I showed my Nexus card at the TSA check *not* realizing it did not match name on ticket. No one blinked and I didn't notice. Forward a few flights months later and I realized they didn't match (to be fair: one is a double barreled name including the other). So I used my old DL that has my current name, matching my ticket, and again no problems with Pre-Check etc. I thought it was all very odd, and would like to get my current legal name on everything but it looks like I cannot for the foreseeable... I am sure you can call me just lucky, but that's at least 8 flights without any issues so far, and 6 different airports.

Amtrak is next and no worries there. :-)
