I don't know about the Tier 1 program, but I do know that they have business development reps that track spend on the business products. At work, we have a business Amex credit card of some kind (it might be one of the Blue products) with about 20 authorized users. I don't know what the spend is, but I'm certain it's well under $1 million a year. One day, an Amex business development rep showed up at our office unannounced to introduce themselves. They knew all of the details about number of cards, spend, and even how many MR points we had, and said that our spend was in the top x percentile of Amex business cards in the area. They were touting other products that we might be interested in, such as business loans. The relevance here is that they definitely have business development people for the business card products, and they will proactively reach out to you at some point once your spend gets their attention.
