No, shouldn't affect it because the $1000 minimum spend is required to trigger the statement credit in the first place. The statement credit is just that a credit to the statement, not a subtraction against purchases. You still made $1K in purchases, fulfilling the requirements.
Statement #1 will close at $1075 and show a balance due of $1075 (including the $75 AF). Your miles and statement credit should post up to a week or so after the statement close. At that point your running balance online will show $975. When Statement #2 closes, it'll show $1075 balance at period open, and credits of $100 statement credit and $975 payment (assuming you've paid in full and not made any additional purchases).
