The math is the math, whether it's multi-line or single line game. A 9/6 JoB game has a house edge of approximately .46% with a relatively low variance. (It's a slightly negative game). That does not change if you play a single line or multi-line game.
I strongly suggest you read up on some video poker strategies. Especially important is learning to play "close to perfect," and understanding the variance of the different games. Variance can be a big issue. Also setting your bankroll properly in relation to the amount you are betting per spin. (Or at least understanding if you are short-bankrolled.)
Broad concepts, most of the "best" video poker in major casino markets are at a high denomination.
