If you are an American citizen, Global Entry is automatically included. If Canadian, they will ask. Then you answer yes or no and you're done. Iris scans are only done in Canada, you will not be able to fly into Canada and use the Nexus line w/o your iris scan. Iris scans do not require an appointment.
For TSA PreCheck, simply entry your known traveler number (you get this when you are approved) into your airline profile. Then you should be all set.
