According to http://en.wikipedia.org/wiki/Non-stop_flight it's SQ21 but I'm starting to question that.
SQ flies IAH-SIN via Moscow (DME), so it's technically 2 flights totaling 23 hours.

EWR-SIN is the longest non-stop flight.
