getting off at an intermediate stop

If you are on a flight that stops somewhere to allow passengers to board and unboard is there anything to stop you getting off at the intermediate stop if you are ticketed through to the final destination? Presumably it would not invalidate the return leg of your flight as the airline wouldn't know you got off early. How do they check that the passengers remaining on the plane are those with luggage in the hold?
