Your status will be updated as soon as the flights post -- usuaully a day or two. UA will credit the 25% RDM bonus on all flights that start after you have achieved the mileage threshold. It can take up to a week for E+ access to show up. The 2P card can take awhile -- 4 to 6 weeks is the typically delay.

As a 2P, TATL upgrades/op-ups are exceptionally rare.
