I doubt that there is a shorter itinerary with only two layovers. But there is a shorter itinerary with four layovers.

e.g.

March 13 3:15-22:50 GRU-DOH QR 774
March 13-14 23:30-1:45 DOH-DXB EK 846
March 14 3:30-14:45 DXB-PEK EK 306
March 14-15 17:25-17:55 PEK-DFW AA 262
March 15 19:10-7:05 AA 963

51 hours and 50 minutes

You can also route through NRT and then DEN or another U.S. gateway before DFW rather than PEK.

Current world record is 52hrs 34min

... as it was achieved last January by an Etihad executive (see here for details).

So actually real people do play this game !

I've tried and 54h45m is the shortest >25,000miles route I can come up with.

1. SYD-SCL QF27 12h20min
2. SCL 1h30min
3. SCL-MAD IB6830 13h40min
4. MAD 3h25min
5. MAD-AUH EY76 6h55min
6. AUH 2h45min
7. AUH-SYD 14h10m

Overall travel time 54h 45min. 47h5min in the air and 7h40min on the ground.
