The file was created using Excel 2003. Excel was set to recalculate formulas automatically, but disabling it didn't seem to make much of a difference. Disabling the antivirus also didn't affect the loading times significantly.
Regarding the first two attempts, first one was a failure and I got impatient with the second one - closed all orphaned instances before trying to open Excel again.
As far as Acrobat, I don't have it running. Using Sumatra PDF instead.

Is it an excel 2007 or previous version file? Is excel configured to recalculate formulas automatically or not? Both of these things can contribute to the speed issue with opening the file. Trying once without AV enabled should pretty easily tell you if that's the problem. Ditto for disabling the automatic formula calculation (formula tab | calculation | automatic).
Also, is Excel already open or not? If not, does opening it up first speed the process notably? There may be issues with add-ins or other things slowing down the overall load process, though probably not 6 minutes worth.
Were the first two attempts actual failures or did you just get impatient? Do you have orphaned instances of excel.exe running?
