I believe they will need to cancel the LX booking and wait for the miles to be redeposited into your account, which will take more than a few seconds... It could take a few days...
However im sure if you do the cancelling and rebooking all in one phonecall and are lucky to get a helpful agent, they may be able to put the new SQ booking onhold for you until the miles are redeposited and you can then call back to complete. But remember SQ bookings can only be held for 24hours so the call centre will need to get the finger out to ensure everything goes smoothly.
