If the travel is purely domestic, there is never any change fee/penalty to upgrade the ticket to a higher fare, provided the flights remain the same.

If the travel is international, it depends on the particular fare rules of the fare you are currently booked on and the fare you upgrade to. Sometimes, there is no upgrade allowed at all, sometimes only to a limited number of fare types, sometimes there is a change fee/penalty, sometimes there isn't.
