In order to cancel your confirmed upgrade, there must be space available in your original booking class (i.e. W,V,Q, etc.). If your original booking class is no longer available, you're pretty much stuck with your upgrade or at the mercy of CSRs and service managers. I'd cancel ASAP, and if that works out, keep looking to see if confirmable upgrades for the long flight become available.
