It'll be a long day if you want to go to the main visitor centre. One of the bottlenecks is getting over the Hoover Dam (bypass bridge won't be completed for another 3 years). Figure at least 8 hours driving each way. As others have suggested, I'd favour allocating more time to visiting the GC rather than in LV.
