Absolutely, and definitely worth the trip! You can take the A3 to Wurzburg (about 100km if I remember correctly), and then take the A7 down directly to Rothenburg. Fun little city. Alternatively, you can take the Romantic Road from Wurzburg through Weikersheim on your way to Rothenburg. Weikersheim has a great castle and very cute town square. Nowhere near as many tourists and it's REAL Germany.
