Looking at the seatmap, to the left of grand staircase there is a toilet, but to the right?

SQ A380 has 471 seats. 747 has 589 seats, on Corsair. 777-300ER has 472 seats on Air France. They only have one story jet bridges - 777 has no upper deck, and 747 upper deck doors are unsuitable for jetbridges.

One jetway? Depends where.

If it is one jetway in first door then obviously all 311 main deck economy passengers must walk through First Class. What do they do on SQ 777-300ER? The 88 upper deck economy passengers have the choice between walking through First Class and rear economy, up spiral stairs and forward again, or else get up the grand staircase, through Business and back to the rear upper deck.

If it is one jetway in second door then the 311 main deck economy passengers simply turn right. The R passengers simply turn left. The C passengers either go through Suites class, up grand staircase and back again, or else straight up the forward lift.

If it is one jetway in upper deck door... then all 323 main deck passengers must get down again. Any airports where this is problem?

Oh, and since jetbridges are forward of wing, there is no way to get to upper deck economy directly. You must either go all the way to the back, then up spiral stairs or rear lift, then forward again, or if you board on upper deck, go back through C class.
