I did a quick run through the Hertz website and Sydney (downtown) to Melbourne (downtown) runs $201 for 3 days ($54.45 per day plus taxes but no drop-off fee). Airport to airport will cost you an extra 18%.
I hired a car through Hertz to drive BNE-MEL a few years ago and they confirmed that they waive the one-way drop-off fee for rental periods of 3 or more days.
