Yes, I have been able to do so, at the gate. I knew that there was space in F so I asked the GA to split the itinerary (I was travelling with a batchelor party mate on an overnight las-bos, wouldn't try dumping my companion when travelling with the wife) so that I could request an upgrade. The GA did so and gave me a new boarding pass showing the V class for medallion upgrades.
I have only used one of these (DL Plat. AMEX) companion certificates.
