What is most important to you, price, speed or comfort? What time of day will it be?

In general, for a first time visitor with a decent amount of luggage, I recommend just hopping in a taxi. It will be a flat fare of $45, plus toll ($4) and tip (customarily 15-20%). At most times of day, this will be a 45-minute trip, but it could range from 30 to 90 minutes depending on traffic.

If you're interested in taking public transportation, you can take the AirTrain from JFK to Jamaica Station ($5) and then transfer to the NYC subway's E train ($2). Take the E train to the Lexington Avenue station and either walk to Grand Central from there (about half a mile down Lexington Avenue) or transfer to the downtown 6 train for one stop. You should plan on this taking 60 to 90 minutes.
