Most of the nwa.com check-in promotions says this in the terms & conditions:
"Promotion applicable for customers who complete the check-in process through nwa.com Check-In or via an E-Service Center for Northwest operated flights only. Changing your reservation after checking in for your flight will result in promotional disqualification"
Accordingly, making manual changes to the seat assignment(i.e changing the flights, stand by for earlier/later flights, rebooking due to flight cancellations etc) after an nwa.com check-in might not fetch you the 5 points. I am not sure how it works if it is a complimentary first class upgrade at the gate though ! Hope you'll still be eligible for the 5 points.
