No, you don't get a second free transfer (bus-subway-subway).

On Thursday evening, I took the M86 crosstown bus from the West Side and transferred for free (as expected) to the downtown 6 train at 86 St/Lexington Av. I exited at 68 St and walked to 63 St/Lexington Av to board the F train, and I was charged a second fare. This probably also means that subway-subway-bus won't work either.

At least in this thread it does. After my last post, I realized that in addition to "Travel Agent" and "Transit Authority", it also often stands for "Ticket Agent" (as opposed to GA or "Gate Agent").
