I'm guessing that the OP plans to take the train to Penn Station. Taking a cab to your hotel really is the best option, as any option involving public transportation will involve a fair bit of walking. If you do want to take public transportation, I agree that the 2 or 3 train to 42nd Street with a transfer to the Shuttle to Grand Central is your best bet.
