The points will likely not post automatically - I don't believe that I've ever had two rooms on the same stay post automatically, even when they were on the same folio. You'll likely have to call or e-mail to get the credit.

Note that this refers to the points only, which is all that you'd get in addition to the points for the first room. The second room will not garner separate stay/night credit.
