It is much much nicer to have them on during the entire flight.
Knowing that my headphones and iPod will have no effect whatsoever on any radio communications, I consider it a personal challenge to see if I can wear my headphones and listen to my iPod all the way through to landing. I'm usually successful about 50% of the time.
