It would not be eligible if you are using an Expedia Special Rate. The terms for using the rate tell you this on the booking page. Also, pre-paid rates to Travelocity would not count either, especially if it is a package deal.
Even if it were eligible, you would have to choose between earning Starpoints or airline miles.
Not sure about the AMEX thing. If the payee is Travelocity or Expedia, probably not. If you are paying the hotel at check-in, then probably.
