Food poisoning is a a frequent occurance on travels to foreign lands. It is a fact of life. If you are not used to travelling, then you should always have your own local suply of medication which your body is used to; rather than using locally available medication which might have other effects on you.

If you were really that concerned about your wife, I am surprised you took time to argue about your bill. At 2 am in the morning, I am not surprised about being charged for the night. It was your choice to seek medical help in Singapore rather locally. If there was an emergency with my partner, I would have just handed over my credit card to be swiped and run out of the hotel. I think it is amusing to be still waited about hand and foot.

I would be concerned about hotel dispensing medication of any kind to guests. Incorrect medication would render the hotel liable to be sued which given the tone of this posting, I am certain you would have no hesitation in doing.

I think it is unreasonable to expect the duty manager to run around you like a lackey when there are other hotel staff around. I do expect the duty manager to turn up when I ask to speak to one and it would appear that he did.

I think it is very important to record complaints on public forums like this to share experiences. The style of communication determines how useful any reasonable reader would find that information.

Regarding the issue about the room not being available, my immediate response would be to point out their check-in policy and ask if they were going to make up for their deficiency in some way (like a drink at the bar or a snack in the restuarant).
