No, they are not likely to get annoyed, unless the one piece is very oversized. Otherwise should not be a problem. Carry-ons normally aren't weighed in SIN.

Depends on how far your gate is. Just make sure you get to the gate at least 10 minutes before the departure time, or they may close it.
