I feel the Q33 takes a lot longer to get to Jackson Heights and that the M60 to Harlem and then the #6 train will be much quicker. Also, the station at Jackson Heights is a bit confusing-- or such had been my experience when I was a newbie in NYC.
On the other hand, even though Jackson Heights does not appear in very many tourist brochures, it is certainly a sight to see which highlights the cultural melting point that NYC is.
Of course, this ultimately depends on when you get in, but for the most part, I'd have to go with taking the M60 to 125th St.; despite the escalators/elevators at 53rd/lex/51st it is still a hike from the E to 6 platform. Also, the Q33 goes southbound from the airport on a one-lane road (82nd St.), which can take some time if there is traffic. (I've actually gotten off a few blocks away from the train station and walked because it was so backed up) The M60 goes down Astoria Blvd., which is a 3-lane road I believe.
As for taking the bus from Grand Central... if it's rush hour, I would go for NJ Transit to Skytrain.
For me, it's always getting to the airport that's half the battle. Getting to my destination on time w/ my checked luggage is the other half.
