Not sure whether Guiness will allow SIN-EWR-SIN or EWR-SIN-EWR. I did have some discussions with them but didnt pursue it.

I can think of other routings that also come in around 48 hours and easily meet distance requirement (think 4 flights on heavy routes each 9+ hours) but 44 hours is going to be tough.
