Benadryl is antihistamine for the treatment for the treatment of allergic reactions, and it is notorious for causing excitability and hyperactivity in children - it says so right on the box! In a small number of children it can also affect their respiratory function.
A few drops of valerian or chamomile (you can buy ones marketed for kids) in a cup/bottle/sippie of orange juice usually will do the trick. The paradoxical excitability that benadryl has on little kids can be QUITE profound (anyone remember the "Cornholio" skits on MTV's Beavis and Butthead? Yeah...just like *that*).
