I once spoke with a GM about a discounted prepaid rate for 150 nights, -- he was fine with the idea and offered me a discounted rate. Basically, the hotel accounts for it as a prepayment/deposit against the ultimate folio.

The OP edited his response to make his intentions a lot clearer, my response was based on a more ambiguous original post.
In which case either of these ideas works to approach a hotel and pre-buy certain nights and agree particular benefits/draw-down concepts. As you correctly say, there are few businesses out there that wouldn't be willing to take a pre-payment of a large chunk of money. Note, that this could also cover multiple people in your company as well, it's just a business negotiation. And primary risk is if the hotel goes out of business, or you no longer need to travel to that city so the money is wasted.
