For starters, if you choose post paid you will not be able to trigger the $200 FHR/THC annual credit that comes with the personal plat card. That only gets triggered with prepaid reservations.
If you select pay at hotel you will definitely get FHR benefits and I'm pretty much positive it will trigger your $100 off $500 offer if your total bill is $500 or more, it doesn't say it has to be only for the actual room charge.
Last year I booked a WA via FHR and selected pay later since I'd already used my $200 plat credit. I received all FHR benefits and paid with my Hilton Aspire that had an Amex Hilton offer saved to it that also said I need to book via Hilton (don't recall the exact $$ amount). The payment triggered that offer as well as the $200 Hilton Aspire Resort Credit. So chances are very high that this will work for you with Peninsula as well.
