Which one would be best based on cost, prefer large sized rooms and bathrooms. Platinum, so good breakfast and lounge is a plus. Ideally good location for us to do the sights of Singapore, with only 2 full days we aren't going to Sentosa.

Below seems to fit within our budget currently:

Aloft Singapore Novena - best rated among all, but rooms on smaller side.

Maxwell/Duxton Reserve - Bad reviews on MB website so not sure if they are good or not?

Courtyard Singapore Novena - Base room on smaller side but rooms look good!

Singapore Marriott Tang Plaza - Large rooms, large bathrooms.

The Westin Singapore - On the expensive side but could still fit in our budget.
The Tang Plaza is probably best for elite benefits. They allow breakfast in either the lounge or the main restaurant. The lounge is spacious and has generous hors d'oeuvres and beer/wine/cocktails in the evening. It is also good central location. Courtyard and Aloft don't have lounge or decent elite benefits. Westin lounge is small and food & beverage selection not as good as Tang Plaza.

Not on your list is the JW Marriott. It sometimes prices very similar to the Tang Plaza and while you cannot take breakfast in the main restaurant, it has a better lounge. The entry level rooms are small though, so you are banking on a upgrade at checkin to get more spacious room.
