I don't expect that you'd be able to up-fare after booking, but an agent should be able to book you the Basic L fare on a new reservation if (a) it exists; (b) they're well-trained; and (c) they're convinced you have a reason to do it.

I concur with the other posters; we're assuming you're trying to tweak mileage earnings in TK Miles and Smiles, and I think this would only work for international BE. Domestically, UA is going to report N no matter what BE fare you purchase.
