yes, but be aware that it can take a few days from the final flight that gets you to 600 TP before Silver appears in your account

Also if I upgrade to business from economy (which im flying) using Avios do I gain tier points? or do I have to pay in cash to earn tier points.

You get TP for cash fares. if you purchase economy using cash you will get economy TP. if you then upgrade with avios you will still get economy TP
